Most likely this is because you are working in Scheduled mode: in this mode content is loaded and played back, based on a pre-defined schedule. All manual actions that could potentially interfere with this schedule are disabled.
In order to use the buttons, you have to go to manual mode. In order to do this, click the "Scheduler on/off" button on the bottom right of the Player module in (Web) Commander.
